History:
First Sailormoon Manga Series (for convenience sake)
She got introduced as the smart girl that didn't talk to anyone. She can be considered the stereotype of Japanese people. (Also a general stereotype in Anime of the "smart shy girl") She was a transfer student that was exceptionally smart, but had no friends. Usagi thought that she was nice, and even though Usagi thought that Ami could help with the homework at first, she quickly saw that Ami was not just smart, but also caring. She could be fun and loosen up too (with a bit of encouragement). She even got a high score on the Sailor V game (which is a measure of how good a Soldier you are according to the manga Code Name wa Sailor V)
In this series Ami is uptight and it even reflects in her character design. She's supposed to be a clean cut student. She's ranking in the top of her class with perfect, or near-perfect scores. She hasn't loosened up yet, so she seems reclusive for most of the series.
She also receives her powers as Sailor Mercury. Sailor Mercury is reflected in her character as well. As Ami starts to loosen up in the later series, so does Sailor Mercury.
Black Moon Manga Series (for convenience sake)
In this series we learn that Sailor Mercury has a special power called dousing. Though it's not followed up in the later series of the manga, it does show her relating more closely to water attacks, and control of water. Ami herself is developed a little bit with the revelation that she is an excellent swimmer! She is also good at chess, which is a strategy game, but none-the-less is a game. This loosens up Ami as just a perfect A student and starts her down the road to being more of a person. (More human at least and less android)
Her character design also loosens too. Her hair becomes longer and more flowing. It seems she is becoming the image of her element. Also I think it was in the second Artbook that she finally got drawn in a bathing suit! ^_^ Also she is much more graceful and doesn't seem as uptight.
Infinity Manga (for convenience sake)
In this series too she starts to loosen up even more, but we don't get to see much of Mercury because this manga mostly focuses on the Senshi from the Outer Solar System. So her character doesn't have many revelations or development. (BTW, the balance of the Senshi from the Outer Solar System and Inners is better in the Dream manga)
The Dream Manga (for convenience sake)
The Dream manga like with the other the Senshi from the Inner Solar System characters made a quite a big development on Ami. This showed Ami as a girl who is very competitive. (This also became apparent in the anime, BTW)
The Stars Manga (for convenience sake)
The Stars manga for the characters was fractured as Takeuchi-sensei decided that more characters was the way to go. Unfortunately this meant that Ami was destroyed by the enemy pretty much right away, for the sake of Usagi's heart showing. So what we learn about Ami is zilch. We do get to see her hair get longer though.
Anime:
The anime expands of Ami's father just a tad more, but her history remains pretty much unchanged. (However, Ami's father in the manga taught her swimming and how to relax).
Personality:
Manga:
She's the brains of the operation. Quiet and graceful in her own way. She is a perfectionist (matches astrology, too). She is very academically strong, though the least powerful of the senshi she uses her brain and analyzing skills to make up for it. She has a rumored IQ of 300, but doesn't always get perfect scores. (299 at least). She loves to study and read. She gets to school 40 minutes early and then reads a book (manga) It's amazing that she gets those high scores with the amount of time taken up as a sailor senshi.
Anime:
She tends to be very insecure in the anime. (Mainly relating to her blood type). They emphasis that she's a perfectionist more, and also that she wants friends. Of course she's no competition for perfection against Michiru, but it does reflect in her personality. There is also a trend in the anime to make Ami seem more like a "normal girl" someone who doesn't just want to become a doctor, but also wants to have a husband and kids. When the girls sneak around she's the one that's always objecting to what they are doing. This aspect in the manga (the girls sneaking around) is not seen.
Hisakawa Aya's performance is really excellent on Ami, it's not a shaky voice at all. It has the flexibility to do all the voicing needed from a bit depressed, to excited, to a bit shy.
NA Dub
Depending on the season and which voice you choose, her character can be different. Karen Bernstein usually interpreted the character as shy and smart. It came across in her voicing. She tended to be able to get her voice to work the parts of Amy that were knowledgeable, without being stiff, and show caring when needed. Though if Ami's First Love was ever aired, I doubt she could do it full justice. (That's very tricky bit of voicing.. and rather suits Hisakawa Aya rather well...). No drastic changes were made to the character, and there seems to be no interference with voice direction (as there was for Usagi to Serena). Thus her character pretty much stays the same.
Lisa Balkman tends to interpret Ami as a bit more stiff, very reserved and insecure. She tends to shake the voice more than Hisakawa Aya did. She tends to have trouble showing Amy's full range of emotion, from concerned to very caring, to excited. She seems to be less confident about the character. I'd say that she probably would not be very good for Ami's First Love Letter at all. I would vote Karen Berstein as the better candidate. Of course, she did not have the full benefit of knowing Amy's full range of character and the subtleties that Amy can present.
Overall, the interpretation for Amy is a bit more flat than that for Ami. Amy lacks the range of emotion that stays in character, and the underlying courage that Hisakawa Aya tends to voice into her character, as well as the quirky humor that Hisakawa-san said she liked to put into her voicing of Ami.

PGSM Live Action:
They made her a lot more shy and reserved. She also has trouble with putting on make-up correctly and is more socially awkward than Ami is in either the manga or the anime. She had to try harder to fit in and also wanted to fit in more badly than in the other versions. This may have been to make her change into Dark Mercury even more dramatic (In PGSM, Mercury becomes an evil senshi for a time).
Why She is So Cool!:
Well, she's drawn better in the manga than the anime. She is a graceful swimmer (her favorite sport) and very reliable. Her hate of love letters makes her a very versatile character. Hey, I would love to be her friend too. She's the favorite of Japan as sited in Takeuchi's liner notes.
